The invention further provides host cells comprising the vectors. The invention further provides methods of using the adenoviral vectors.

Replication-competent adenoviral vectors comprising an EBV-specific transcriptional regulatory element (TRE) operably linked to a gene required for adenovirus replication are provided. By providing for transcriptional initiating regulation dependent upon transcription factors that are only active in specific, limited cell types, virus replication can be restricted to particular target cells. The modified adenovirus may be used as a vehicle for introducing new genetic capability, particularly associated with cytotoxicity for treating neoplasia.

Method For The Purification, Production And Formulation Of Oncolytic Adenoviruses
A process is provided for the production of substantially pure replication competent adenovirus, together with improved formulations for maintenance of infectivity following storage. In the process, virus infected cells are lysed with detergent. An initial purification step utilizes a pass through a high throughput ion exchange filter. The eluant is treated with nuclease, then refiltered on a high throughput ion exchange filter. The virus formulation provides for enhanced stability of liquid viral preparations at 5 C.
